torCp based Trimethylamine N-Oxide sensor
This composite part links the torC promoter to a GFP (BBa_K142900) reporter with a strong RBS (BBa_M5008) combined with an ampicillin selection marker.
Usage and Biology
This part has been designed for K-12 Escherichia coli and other lab strains of E. coli that have the torCAD operon. This part should allow GFP to be produced in elevated levels in response to trimethylamine N-oxide (TMAO). If TMAO is present, it can be sensed by binding to the torT protein (a periplasmic TMAO-binding protein), which then interacts with the membrane protein TorS to form a sensory complex, altering the behavior of the regulator protein torR. This activated torR protein acts to then transcribe a gene torC. This sequence of events allows E. coli to reduce TMAO in the absence of oxygen in order to perform respiration. Therefore, in the presence of TMAO, E. coli will signal metabolic changes from fermentation to anaerobic respiration by activating torR, making the operon of the gene torC of interest in this device.
